Lactoferrin Decreases Monocyte - Induced Fibroblast Production of Myeloid Colony - Stimulating Activity by Suppressing Monocyte Release

چکیده
Lactoferrin (If) is a negative regulator of myelopoiesis which operates by suppressing the release from mononuclear phagocytes of GM colony-stimulating factor (GMCSF) or monokines which can then induce the release of GM-CSA from accessory cells. In this study. endotoxindepleted. purified iron-saturated human If was assessed for its effect on the production of interleukin-1 by cultured monocytes and their subsequent effect on colony-stimulating factor release from cultured fibroblasts. Monocytes were grown with or without If and If that had previously been incubated with monoclonal anti-If. The monocyteconditioned medium was then either assayed for the presonce of interleukin-1 (Il-i ) with an enzyme-linked immunosorbent assay or for its ability to stimulate fibroblasts to release growth factors for CFU-GM. BFU-E. or CFU-Mix colonies. In the presence of If (iO or i0 mol/I). GM colony-stimulating activity (GM-CSA) was suppressed by

متن کاملRegulation of colony-stimulating activity production. Interactions of fibroblasts, mononuclear phagocytes, and lactoferrin.
Neonatal skin fibroblasts were cultured in supernatants of peripheral blood monocytes that had been cultured with and without lactoferrin. Granulocyte-monocyte colony-stimulating activity (CSA) was measured in supernatants of the fibroblast cultures with normal T lymphocyte-depleted, phagocyte-depleted, low density bone marrow target cells in colony growth (colony-forming unit granulocyte/macro...

Human umbilical vein endothelial cells were cultured in supernatants of peripheral blood monocytes that had been cultured for 3 days with and without lactoferrin. Colony-stimulating activity (CSA) was measured in supernatants of the endothelial cell cultures and appropriate control cultures using normal, T-lymphocyte-depleted, phagocyte-depleted, low-density bone marrow cells in colony growth (...
متن کاملInterleukin 1 stimulates fibroblasts to produce granulocyte-macrophage colony-stimulating activity and prostaglandin E2.
Granulocyte-macrophage colony-stimulating activity (GM-CSA) can be produced by a variety of normal cell types including mononuclear phagocytes, activated T lymphocytes, endothelial cells, and fibroblasts. Recent evidence shows that a major role of the monocyte-macrophage is the recruitment of environmental cells, i.e., fibroblasts, to produce GM-CSA. In this study we have identified interleukin...
